    Cyclic fatigue damage characteristics observed for simple loadings extended to multiaxial life prediction

    Fully reversed uniaxial strain controlled fatigue tests were performed on smooth cylindrical specimens made of 304 stainless steel. Fatigue life data and cracking observations for uniaxial tests were compared with life data and cracking behavior observed in fully reversed torsional tests. It was determined that the product of maximum principle strain amplitude and maximum principle stress provided the best correlation of fatigue lives for these two loading conditions. Implementation of this parameter is in agreement with observed physical damage and it accounts for the variation of stress-strain response, which is unique to specific loading conditions. Biaxial fatigue tests were conducted on tubular specimens employing both in-phase and out-of-phase tension torsion cyclic strain paths. Cracking observations indicated that the physical damage which occurred in the biaxial tests was similar to the damage observed in uniaxial and torsional tests. The Smith, Watson, and Topper parameter was then extended to predict the fatigue lives resulting from the more complex loading conditions

    Evaluating the Economic Impact of Countervailing Duties on United States Warm Water Shrimp Imports

    Estimates of price and scale elasticities for U.S. consumed shrimp are derived using aggregate source country shrimp import data. It was assumed that supply was perfectly elastic and U.S. wholesalers determine the quantities imported from individual countries given the prices and preferences of U.S. consumers. Ex-ante analysis suggests that most countries levied with the countervailing duty experience declines in U.S. import demand while those countries not affected by the countervailing duty experience increases in import demand. Ex-post analysis shows the reverse to be true. Several countries impacted by the countervailing duty had increased import demand from the United States while Mexico, which was not affected by the countervailing duty, had decreased import demand. The results from aggregate level data suggest that imposing duties on specific companies within a country may not be effective if that company is not a monopoly producer or controls a significant share of the shrimp produced in that country.International Relations/Trade,

    Using a fuzzy inference system to control a pumped storage hydro plant

    The paper discusses the development of a fuzzy inference system (FIS) based governor control for a pumped storage hydroelectric plant. The First Hydro Company's plant at Dinorwig in North Wales is the largest of its kind in Europe and is mainly used for frequency control of the UK electrical grid. In previous investigations, a detailed model of the plant was developed using MATLAB(R)/SIMULINK(R) and this is now being used to compare FIS governor operation with the proportional-integral-derivative (PID) controller currently used. The paper describes the development of an FIS governor, and shows that its response to a step increase in load is superior to the PID under certain conditions of load. The paper proceeds to discuss the implications of these results in view of the possible practical application of an FIS governor at the Dinorwig plant

    Defining stage-specific activity of potent new inhibitors of Cryptosporidium parvum growth in vitro

    Currently, nitazoxanide is the only FDA-approved treatment for cryptosporidiosis; unfortunately, it is ineffective in immunocompromised patients, has varied efficacy in immunocompetent individuals, and is not approved in infants under 1 year of age. Identifying new inhibitors for the treatment of cryptosporidiosis requires standardized and quantifiable in vitro assays for assessing potency, selectivity, timing of activity, and reversibility. Here, we provide new protocols for defining which stages of the life cycle are susceptible to four highly active compound classes that likely inhibit different targets in the parasite. We also utilize a newly developed long-term culture system to define assays for monitoring reversibility as a means of defining cidal activity as a function of concentration and time of treatment. These assays should provide valuable in vitro parameters to establish conditions for efficacious in vivo treatment.Cryptosporidium parvum and Cryptosporidium hominis have emerged as major enteric pathogens of infants in the developing world, in addition to their known importance in immunocompromised adults. Although there has been recent progress in identifying new small molecules that inhibit Cryptosporidium sp. growth in vitro or in animal models, we lack information about their mechanism of action, potency across the life cycle, and cidal versus static activities. Here, we explored four potent classes of compounds that include inhibitors that likely target phosphatidylinositol 4 kinase (PI4K), phenylalanine-tRNA synthetase (PheRS), and several potent inhibitors with unknown mechanisms of action. We utilized monoclonal antibodies and gene expression probes for staging life cycle development to define the timing of when inhibitors were active during the life cycle of Cryptosporidium parvum grown in vitro. These different classes of inhibitors targeted different stages of the life cycle, including compounds that blocked replication (PheRS inhibitors), prevented the segmentation of daughter cells and thus blocked egress (PI4K inhibitors), or affected sexual-stage development (a piperazine compound of unknown mechanism). Long-term cultivation of C. parvum in epithelial cell monolayers derived from intestinal stem cells was used to distinguish between cidal and static activities based on the ability of parasites to recover from treatment. Collectively, these approaches should aid in identifying mechanisms of action and for designing in vivo efficacy studies based on time-dependent concentrations needed to achieve cidal activity

    Transforming a 4th year Modern Optics Course Using a Deliberate Practice Framework

    We present a study of active learning pedagogies in an upper division physics course. This work was guided by the principle of deliberate practice for the development of expertise, and this principle was used in the design of the materials and the orchestration of the classroom activities of the students. We present our process for efficiently converting a traditional lecture course based on instructor notes into activities for such a course with active learning methods. Ninety percent of the same material was covered and scores on common exam problems showed a 15 % improvement with an effect size greater than 1 after the transformation. We observe that the improvement and the associated effect size is sustained after handing off the materials to a second instructor. Because the improvement on exam questions was independent of specific problem topics and because the material tested was so mathematically advanced and broad (including linear algebra, Fourier Transforms, partial differential equations, vector calculus), we expect the transformation process could be applied to most upper division physics courses having a similar mathematical base.Comment: 31 page

    Version 4 of the CRU TS monthly high-resolution gridded multivariate climate dataset

    CRU TS (Climatic Research Unit gridded Time Series) is a widely used climate dataset on a 0.5 degrees latitude by 0.5 degrees longitude grid over all land domains of the world except Antarctica. It is derived by the interpolation of monthly climate anomalies from extensive networks of weather station observations. Here we describe the construction of a major new version, CRU TS v4. It is updated to span 1901-2018 by the inclusion of additional station observations, and it will be updated annually. The interpolation process has been changed to use angular-distance weighting (ADW), and the production of secondary variables has been revised to better suit this approach. This implementation of ADW provides improved traceability between each gridded value and the input observations, and allows more informative diagnostics that dataset users can utilise to assess how dataset quality might vary geographically
